Abstract Introduction. Pelargonium graveolens is a well-known ethnomedicinal plant used in traditional medicine. However, Pelargonium zonale has not been investigated thoroughly. Objective. The present study is designed to validate a new HPLC method according to ICH guidelines for pharmaceutical products, and evaluating antimicrobial activity for plants extract. Material and methods. The aerial parts were extracted successively with solvents with different polarities. Antioxidant activity was confirmed by measuring the free radical scavenging activity of 2,2-diphenyl-2-picrylhydrazyl. An Agilent C18 with a gradient elution program using a mixture of acetic acid added to 1% water (A) and methanol (B) was performed for chromatographic separation, and disc diffusion method was used to determine the antimicrobial activity. Results. The method was successfully applied to the analysis and separation of 12 polyphenols compounds. The total phenol content measured by the Folin-Ciocalteu assay method ranges from 159.43 to 164.25 mg GAE/g, and the total flavonoids range from 26.92 to 63.68 mg QE/g. Furthere, extracts showed a moderate to strong antimicrobial activity against five strains with MIC ranging from 0.46 to 3.9 mg/mL. Conclusion. The developed method was accurate, precise, rapid, and sensitive enough for the simultaneous quantitative determination of 12 compounds in our extracts. Additionally, the HPLC-DAD results were strongly correlated with the results found by the assays and confirm the richness of the extracts by the phenolic compounds.
